9 Section 2 General Information 2.1 Safety information Please read this entire manual befo re unpacking, setting up o r operating this equipment. Pay attention to all danger , warning and caution statem ents. Failure to do so cou ld result in serious injury to the operator or damage to th e equipment.
10 General Information 2.1.3 Class 1 LASE R A Class 1 LASER is installed in this instrument. Class 1 LASERS are produc ts where the rad iant power of the LAS ER beam accessible (the accessible emissi on) is always below the Maximum Permissible Exposure value.
11 General Information 2.2 Overview of product The DR 5000 S pectrophotometer is a complete scanning UV/VIS spectrophotometer with a wavelength r ange of 190 to 1 100 nm. The instrument comes with a complete set of application programs a nd multi-language support.

13 Section 3 Inst allation WA R N I N G Electrical and F ire Hazards. Use only the pr ovided power supply . Only qualified per sonnel should conduct the tasks described in this sect ion of the manual.
14 Inst allation 3.3 Connection The DR 5000 has three USB interf aces as a standard feature, located on the back of the instrument ( Figure 1 ). The USB T ype A interfaces are used for communications with a printer , USB memory stick or ke yboard. A USB memory stic k is used to update instrument sof tware.
15 Inst allation 3.4 Cell comp artment, Multi Cell Ho lder , 100 mm Cell Adapter (optional) The DR 5000 S pectr ophotometer comes equippe d with a Multi Cell Holder ( Figure 3 on page 16 ) which is the S tandard Holder , provided with each instrume nt .

19 Section 4 S t a rt Up 4.1 Power the ins trument on and off 1. Plug in power supply . 2. Close the empty cell compartment. 3. T urn the instrument on by pressing the power switch on the back. Note: Do not turn the instrument off an d on in rapid succession.

21 Section 5 S t andard Operations 5.1 Overview 5.1.1 Tip s for the use of the touch screen The entire screen is to uch-activate d. T o make a se lection, pres s the screen with a fingernail, fingertip, pencil eraser or a stylus. Do not press the screen with a sharp object, such as the tip of a ball point pen.
22 St an dard Operations 5.1.3 Main Menu A variety of modes may be selected fr om the Main Menu. The following table br iefly describes each menu option.
23 St andard Operations 5.2 Instrument Setup mode 1. Select Instrument Setup in the Main Menu. A selection of functions appears in order to configure the functions of the instrument. This display appears when the mult i-cell holder is installed or the 13 mm round cuvette/vial comp artmen t is use d.
24 St an dard Operations 5.2.2 Sample ID Use this option to ente r up to 30 Sample Identification tags (up to 13 character s each) into th e instru ment. Sample IDs can be used to specify the sample location or ot her sample specif ic information. 1. Press Sample ID in the Instrument Setup.
25 St andard Operations There are two ways to enter or change a Sample ID in the Measurement Mode: Option 1 In the result display , press Options > More > Instrument Setup . If a Sample ID is already assigned, sele ct the Sample ID symbol in the result display .
26 St an dard Operations 2. Select Long to change the number of audio signa ls. Use the alphanumeric keyp ad to enter/specify the number of audio sign als (4–25). Note: A high number of audio signals increases the duratio n of the tones and a small number of audio signals reduces the duration o f the tones.
27 St andard Operations 5. Select the length of time the lamp will be switched on. Note: After this period of time the lamp automatically turns off, after no measurement has been made o utside the corresponding range (UV/VIS). 6. T ouch OK to confirm.
28 St an dard Operations When the lamp is warmed up and re ady , the UV icon stops flashing. Note: If both lamps are On, the icon UV -VIS is displayed in the selected Measurement Mode. Note: Alternatively you can touch the UV- or VIS-Lamp icon on the measurement screen to change the actu al setting.
29 St andard Operations 5.2.6 PC and printer The instrument is provided with 3 USB interfaces, which are located on the back of the instrument (see Figure 1 on page 14 ). These interfaces c an be used for expor ting data and graphics to a pr inter , updating dat a and for dat a communication to a personal computer .
30 St an dard Operations Printer Setup: • Resolution: Print quality • Paper: Paper size Note: If an optional Thermal Pri nter is connected, the function "Auto Send" on/off is available. 4. Select Auto-Send: On to send all measured dat a automatically to the Thermal printer .
31 St andard Operations 7. Press Paper to select the paper size. Select between • Letter • Legal • Executive • A4 8. Press OK to confirm. Note: Press OK again to return to the PC & Printer menu. 5.2.6.2 Print dat a 1. Press Recall Dat a in the Main Menu.
32 St an dard Operations 4. Select Auto-Send: On to send all measured dat a automatically to the PC. Note: If Auto-Send: Off is selected, the PC & Printer icon must be pressed, in order to send data to the PC. Note: The remote funct ion is onl y fo r monitoring the data transfer .
33 St andard Operations 4. Press Security List to lock various functions for unauthorized users. 5. Highlight the d esired funct ions to cont rol. 6. Confirm the Security List wi th OK to return to the Password menu. 7. Press On to highlight the new settings of the Security List.
34 St an dard Operations 3. Press Off to deactivate the settings of the Security List. 4. Press OK to return to Instrument Setup. Note: Use this function to dele te the former Password or to enter a new one.
35 St andard Operations 5.3 S tore, recall, send and delete dat a 5.3.1 The Dat a Log The Data Log will store up to 1000 readings taken in the modes: S tored Pro grams, User Programs, Favorite Programs, Single W avelength and Multi W avelength .
36 St an dard Operations 4. The function Filter Settings is used to search for specific items. 5. Highlight On to turn on the filt ers to select data by • Sample ID • Operator ID • St a r t D a t e • Parameter or any combination of the four . 6.
37 St andard Operations 5.3.1.4 Delete stored dat a from the dat a log 1. Press Recall Dat a in the Main Menu. 2. Press Dat a Log>Options>Delet e . 3. Highlight Single Po int or Filtered data or All dat a and press OK to confirm. Note: The number in parentheses is the total number of data set s assigned to this selection.
38 St an dard Operations 5.3.2 S tore, recall, send and delete dat a from wavelength scan and time course The instrument can store 20 W avelength Scans and 20 T ime Course Data sets.The dat a can be stored manually at the user's discretion after viewing the da ta.

42 St an dard Operations 5.4 S tored Programs The instrument contains more than 200 programmed procedures. They can be accessed th rough the St ored Programs menu. 5.4.1 Select a saved test/method; enter user-specific basic dat a 4. Press Stored Programs in the Main Menu to view an alphabetical list of stored programs with pr ogram numbers.
43 St andard Operations Store Off/On With the Store On setting, all measu rement data ar e stored automatically . With the Sto r e O ff setting, no measurement data are stored.
44 St an dard Operations 5.4.3 Use of program timers Some procedures do not r equire the use of timers. Other procedures require se veral timers. T hese timers are pre-progr ammed into e ach Stored Program , alo ng with a description of the activity to be performed durin g the timed period.
45 St andard Operations 5.4.4.1 T ake single wavelength measurement s (single reading) 1. Insert the blank cuve tt e into the cuvette/ sample cell holder. Press Zero . Note: The Read key is only active after the zero measurement has been carried out. 2.

48 St an dard Operations 5.4.8 Run a reagent blank Some of the stored test s/meth ods include the "Reagent Blank" function. This enables the reagen t blank value to be measur ed and then taken into accoun t in calculating the measurement result.
49 St andard Operations 5.4.9 Analysis of samples 1. Press S tored Programs and select a progra m. 2. Insert the blank cuvette/cell into the cuvette/sample cell holder . 3. Press Zero . 4. Remove blank cuvette/cell and in sert sample cuvette/cell into the cell compartment.
50 St an dard Operations 5.5 Barcode Programs A special barcode reader in ce ll comp artment #1 automatically reads the barcode on the 13 mm cu vette/vial as the cuvette/vial completes a single rot ation.
51 St andard Operations 5.5.1 Complete a barc ode 13 mm test/vial Note: The cell compartment must be closed before a mea surement is made! 1. The instrument needs to be at the Main Menu.
52 St an dard Operations 1. In the result display , press Options>More>Chemical Form . A list of available evaluation fo rms appears. 2. Select the required chemical form an d press OK to confirm. Note: The selected chemical fo rm is displayed, but does not become the default.
53 St andard Operations 5.5.5 Sample blank T urbidity and color in t he sample matrix can fa lsify the results of a photometric analysis. The interference factors come from the sample or are cr eated by reactions with the reagents. The influence of tur bidity and/or color can be eliminated or reduced by taking a sample blank reading.
54 St an dard Operations procedure. Other barcod e tests use the reagent test vials to determine the sample bla nk. Please refer to the Hach procedure for method-spe cific instructions.
55 St andard Operations Enter the V ersion 1. Highlight the line cont aining V ersion and press Edit . Use the alphanumeric keyp ad to enter the name indica ted in the working procedure. 2. Press OK to confirm. Enter the Measurement Process For detaile d information on enterin g the measurement process, see section 6.
56 St an dard Operations Enter the V ariables (Factors, W avelength and Conversion Factors) For det ailed information on entering the V ariables, see section 6.1.2 on page 72 . Check the working pr ocedure in adva nce to determin e whether this point needs to be cha nged.
57 St andard Operations 5.5.6.2 Up date an existing barc ode test Note: Only in very few cases does the re vision of a test require all test specifications to b e updated. Option 1: Manual up date of test data 1. The instrument needs to be at the Main Menu.
58 St an dard Operations 5.5.6.3 Program a new test 1. The instrument need s to be at the Main Menu. 2. Insert the sample cuve tte/vial or blank (depending on the working procedure) into th e cell compartm ent #1 and close the cell compartment.

61 Section 6 Advanced Operations 6.1 User Programs User programs provide the op portunity to complete "made to measure" analysis. The User Progra ms database is em pty when the instrum ent leaves the factory and is used to acco mmodate programs cre ated by users specific need s.
62 Advanced Operations Program Numb er: S pecific test number , with whic h the progra m can subseq uently be called up from the selection list in the User Programs menu or the Favorites menu. 2. Use the alphanumeric keyp ad to enter a program number between 950 and 999.
63 Advanced Operations 6.1.1.1 Single wavelength settings If the Single Wavelength m ode is selected, the following parame ters can be defined: Unit s: Select the required unit from the list and press Next . Note: Units of measure not included in this list can be added in the edit program under Program Options , Edit .
64 Advanced Operations Chemical form: Enter the chemical formula used in the display to represent the analysis parameter . Use the alphanumeric keyp ad to enter the chemical form and press Next to enter Calibration settings.
65 Advanced Operations List of available absorbance for mulas A 1 is the absorbance at waveleng th 1, A 2 is the absorbance at waveleng th 2 and so on K 1 is the factor at wavelength 1, K 2 is the factor at wavelength 2 and so on If a subtraction has to be completed, the factor s can be entered with a minus sign.
66 Advanced Operations Chemical form: Enter the chemical formula used in the display to represent the analysis parameter . Use the alphanumeric keyp ad to enter the chemical form and press Next to enter Calibration settings.
67 Advanced Operations 2. T o enter the standard concentrations and cor responding absorbance value s in the displayed t able, press the " + " symbol. Use the alphanume ric keypad to enter the values. Press OK and enter th e corresponding absorbance value.
68 Advanced Operations Calibration by r eading st andards 1. Press Read St andards and press Next . 2. T o enter the standard con centrations in the displa yed table, press the " + " symbol. Use the alphanumeric keyp ad to enter the standa rd concentration.
69 Advanced Operations 9. Press Force 0 to change the setting from Of f to On . The curve then passes throug h the origin of the coordinate system. Note: This may have an adverse effect on the correlation coefficient (r 2 ). 10. Press Ta b l e to display the table again.
70 Advanced Operations 6.1.1.4 Store a user program The input of the basic data is complete. An overview of the variable program d ata is displayed. 4. T o enter more specifications or change existing ones, highlight the appropriate line and press Edit .
71 Advanced Operations Timer 1 / T imer 2 / Timer 3 / T imer 4: This function can define time interv als for up t o four timers. T imer designations such as Shake, W ait and Swirl can be assigned. 1. Highlight the appropriate line in the overview of the program data an d press Edit .
72 Advanced Operations 6.1.2 Free programming program type Free Programming is an advan ced option for entering original user-devel oped methods. When the Free Programming option is selected, an overview of the specifications of the progra mmed test is displayed.
73 Advanced Operations • Should individual program seq uences be repeate d? The element s of a measuring sequence, such as zero and sample measurement s and the timer(s) (reaction ti mes, waiting times, etc.
74 Advanced Operations Process Ti mer key 1. Press the Process Timer key to enter any wait ing, reaction or handling tim es that have t o be taken into acc ount. Use th e alphanumeric keyp ad to enter the time. Press OK and co nfirm the input by pressing OK again.
75 Advanced Operations Delete an element of a measuring sequence Select the appropriate line and press Delete . The element is deleted. Insert an element of a measur ing sequence Select the line in the mea suring sequence where the insertion is be made and press New .
76 Advanced Operations Refer to Table 11 for detailed information on the Edit formula keys. Note: The evaluation formula is built up successively in the disp lay in accordance with the input. Note: The arrow key del etes the most recent ly entered element of the formula.
77 Advanced Operations Enter the ne xt calibration formula (C2 or Cn) 5. Highlight C2: Off a nd press Edit . 6. Select C2: Off again and press Edit . The display switches to C2: On . 7. Highlight the next line C2 = to define the formu la and press Edit .
78 Advanced Operations Select Conc. key If an already defined formula, in this case C1, is to be take n into account in the formula for C2, p ress Select conc. Enter the nu mber of th e formula (e.g. 1 for C1 ) and pres s OK to confirm. Cn can now be linked with a mathematical operation.
79 Advanced Operations 6.1.3 Select a user program 1. Press User Programs in th e Main Menu to view an alphabetical list of user pr ograms with progra m numbers. The User Programs list will appear . Note: Use the scroll bar to scroll throug h the list quickly .
80 Advanced Operations 6.1.4.1 Add to Favorites 1. Press Add to Favorites and press OK to confirm. The program is ad ded to the Favorites. 6.1.4.2 Edit 1. Press Edit and pres s OK to confirm. An overview of the specificati ons of the programm ed test is displayed.
81 Advanced Operations 6.2 Favorite Programs The most frequently used tests/methods in the St ored Programs menu and the User Programs menu can also be added to the list of favorites to simp lify their selection. T o add Stored Programs and/or User Programs to the favorites list or the favorite prog rams, see section 6.
82 Advanced Operations 6.3 S t andard Addition – monitoring/check ing result s The accuracy of measured values (their correspondence with the actual concen tration of the analyte in the sample) and .

87 Advanced Operations 6.4 Single W a velength (absorbance, concentration and tran smitt ance measurement s) The Single W avelength mode can be used in thr ee ways. For sample measurement s at a single wavelength, the instr ument can be programmed to measure the absorba nce, % transmittance or concentration of the analyte .
88 Advanced Operations Concentration factor: 1. Press Concentration Factor: Off in the Options m enu. Press On to highlight this feature. 2. Press the ”Factor“ key and use the alphan umeric keypa d to enter the f actor by w hich absorban ce reading s are to be multiplied.

90 Advanced Operations 6.5 Multi-W avelength mode – measureme nt s at more than one wavelength In the multi-wavelength mode, absorbance values can be measured at up to four wavelengths and the result s can be mathematically processed to obt ain sums, dif ferences an d relationship s.
91 Advanced Operations λ / Absorbance formula: 1. Press Absorbance For mula . 2. The formula selected in the to p key determines the number of wavelength and coef ficent keys that will appear below . T o change the absorbance formula, pr ess the top key , select a formula from the displayed list and press OK .
92 Advanced Operations 3. T o change a wavelength, press one o f the ” λ x:“ keys. Enter the desired wavelength coef ficient into the numeric keypad . Press OK to confirm. 4. T o change a coefficient, press one of the ”K X :“ keys. Enter the desired coef ficient into the numeric keypad.
93 Advanced Operations 6.5.2 Complete a measurement in the multi wavelength mode 1. Insert the blank cuvette /cell into the cuvette/sample cell holder . Press Zero . Note: The Read key does not be come active until th e zero measurement has been completed.
94 Advanced Operations 6.6 W avelength Scan mode – recordin g of absorbance and transmittance spectrums In the wavelength scan mode, the absor bance of the light in a solution over a defined wavelength spectrum is measured. The measurement result s can be displayed as a curve, as percent transmitta nce (%T) or as absorbance (Abs).
95 Advanced Operations λ Setting wavelength 1. Press the λ key in the Options menu to select the wavelength range and the wavelen gth step. 2. Press the upper lef t key to open the numer ic keypad and select the minimum wave length. Press OK to confirm.
96 Advanced Operations 5. According to the selected wavelength range, select the interval from the actively displayed wa velength steps. In summery maximally 780 measuring step s can be accomplished during a Scan.
97 Advanced Operations 4. Press Return to return to the scan mode. Note: The Integral is shown instead of the date on the disp lay . Note: For the next scan measurement the setting for the Integral will be On . Scale & units 1. Press Scale & Unit s .

99 Advanced Operations 6.6.3 Work with reference scans There are two options to wo rk with Reference Scan : Option 1: 1. Press Reference: Off in the Options menu to select another scan to display on the same sc reen with the current scan. Highlight the required scan number and press Highlight Reference .
100 Advanced Operations Option 2: 1. Insert the blank cuvette/cell into the cuvette/sample cell holder . Press Zero . 2. Insert the sample cuve tte/cell into the cuvette/sample cell holder . Press Read . • The newly plotted wa velength scan curves are sh own in black.
101 Advanced Operations 6.7 T ime course of ab sorbance/transmitt ance The T ime Course Mode is used to collect dat a in either absorbance or transmitt ance for a user-specified length of ti me. After th e data are collected, they can be displayed in eithe r graphic or tabular format.
102 Advanced Operations Time & interval: 1. Press Time & Interval in the Options menu. 2. Input the total time an d the reading time and press OK to confirm.
103 Advanced Operations • Select Mark to mark the next data po int collected. This mark is not used by the instrument, but is available for the user and may indicate a significant even t, such as the additio n of a sample or other reagent. Th e mark is also shown in the table.
104 Advanced Operations 6.7.3.1 Navigation of a time scan or a time scan analysis After a time sca n has been completed, the time and the absorbance/transmitt ance dat a are displayed as a curve. Where the cursor is positioned on the curve, the elap sed time up to this point and the corresponding abso rbance are highlighted.
105 Advanced Operations 6.8 System checks 1. Press System Checks in the Main Menu. The System Checks menu contain s instrument information an d various performance te sts. 6.8.1 Instrument information 1. Press Instrument Information in the System Checks menu.
106 Advanced Operations 6.8.2 Upgrade of the instrument sof tware T o obtain the sof tware for the update from the Intern et at www .hach.com : 1. Go to http://www.hach.com . 2. On the DR 5000 product page, click Lab System Sof tware / Sof tware Up date Downloads u nder the heading Downloads .
107 Advanced Operations 6.8.3.1 W avelength check The W avelength Check test is used to check wavele ngth accuracy at 656.1 and 486.0 nm and to check bandwidth at 656.1 nm, only . 1. Press W avelength Check in the Optical Checks menu. 2. Press λ to select the wavelength.
108 Advanced Operations 5. The result is displayed. 6. Press Cancel to return to Optical Checks. 6.8.3.2 Noise check The Noise Check test is used to test the photometric noise in the instrument. This test can be used to test noi se at any wavelength and at an absorbance level determined by a specific sample.

Thir ty readings ar e averaged for the blank. The ”A verage“ and ”S td Deviation“ are calculated from 100 consecutive absorbanc e re adings (Noise, Sample, Reference) .
11 0 Advanced Operations 6.8.3.3 Stra y light check The S tray Light t est is used t o measure the stray lig ht in the instrum ent at 220.0 nm an d 340.0 nm. 1. Press Str a y Li g ht C he c k in the Optical Checks menu. 2. Press λ to select the wavelength.
111 Advanced Operations 6. The ”Average“ an d ”S td Deviation“ are ca lculated from 100 successive abso rbance m easurem ents. The r esult is displayed and is to be comp ared with the nominal/stan dard data (given in the quality control certificate) of the sample cell/test filter .
